In the movie the biggest lie is belief in God. In a society where everyone tells the truth, the main character hits upon the idea of lying. According to this review, the rest of the movie is about the "man in the sky" lie used to launch into a full on attack on religion and Christians.

"Invention of Lying" -Anti-Religious, Anti-Christian, Pro-Atheist Movie?
Initial reports on this movie is that it's a pro-atheist anti-religious movie. The trailers show it as a cute comedy. The director, Ricky Gervais, is reportedly a militant atheist. Reviewer Kyle Smith says the movie "is a full-on attack on religion in general and Christianity in particular. It might be the most blatantly, one-sidedly atheist movie ever released by a major studio, in this case Warner Bros."
